Background: Secreted phospholipase A2 type IIA (sPLA2-IIA) is a well-established pro-inflammatory protein and has been a major target for drug discovery. Our recent studies found that sPLA2-IIA activates integrins in an allosteric manner through direct binding to a newly identified binding site of integrins (site 2), which is distinct from the classical RGD-binding site (site 1). The sPLA2-IIA-induced integrin activation may be related to the signaling action of sPLA2-IIA.
Principle of the Assay: Mouse Secreted Phospholipase A2 (sPLA2) ELISA Kit employs a two-site sandwich ELISA to quantitate sPLA2 in samples. An antibody specific for sPLA2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any sPLA2 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Mouse sPLA2 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of sPLA2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
