Background: Thymosin-beta-10 is closely related in sequence to TMSB4 and is also an actin-sequestering protein. In most tissues of the rat, the same molecular weight species of TMSB10 mRNA is found; This finding, together with primer extension experiments, suggested that the 2 mRNAs are transcribed from the same gene through a combination of differential promoter utilization and alternative splicing. Both mRNAs were present in pachytene spermatocytes; only the testis-specific mRNA was detected in postmeiotic haploid spermatids. Immunohistochemical analysis showed that the protein was present in differentiating spermatids, suggesting that the testis-specific TMSB10 mRNA is translated in haploid male germ cells. thymosin-beta-10 protein synthesized in adult testis was identical in size to that synthesized in brain.

An antibody specific for TMSB10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TMSB10 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Mouse TMSB10 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of TMSB10 bound in the initial step.
