Background: Troponin T (TnT) is a central player in the calcium regulation of actin thin filament function and is essential for the contraction of striated muscles. Three homologous genes have evolved in vertebrates to encode three muscle type-specific TnT isoforms: TNNT1 for slow skeletal muscle TnT, TNNT2 for cardiac muscle TnT, and TNNT3 for fast skeletal muscle TnT.
Principle of the Assay: Mouse Troponin T, fast skeletal muscle (TNNT3) ELISA Kit employs a two-site sandwich ELISA to quantitate TNNT3 in samples. An antibody specific for TNNT3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TNNT3 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Mouse TNNT3 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of TNNT3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
