Principle of the Assay||The Human Mesothelin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Mesothelin in Cell Culture Supernatants, Serum, Plasma, Urine. This assay employs an antibody specific for Human Mesothelin coated on a 96-well plate. Standards and samples are pipetted into the wells and Mesothelin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Mesothelin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Mesothelin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background/Introduction: Mesothelin, also known as MSLN or MPF is a protein that in humans is encoded by the MSLN gene. This gene is mapped to 16p13.3. This gene encodes a precursor protein that is cleaved into two products, megakaryocyte potentiating factor and mesothelin. Megakaryocyte potentiation factor functions as a cytokine that can stimulate colony formation in bone marrow megakaryocytes. Mesothelian is a glycosylphosphatidylinositol-anchored cell-surface protein that may function as a cell adhesion protein. This protein is overexpressed in epithelial mesotheliomas, ovarian cancers and in specific squamous cell carcinomas.

Product Notes
The Human MSLN msln (Catalog #AAA261673) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA261673 ELISA Kit recognizes Human MSLN. AAA Biotech's Mesothelin can be used in a range of immunoassay formats including, but not limited to, ELISA (Sandwich ELISA). Researchers should empirically determine the suitability of the MSLN msln for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Mesothelin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
