(Cathepsin B staining in Jurkat T cells after treatment with inhibitors.Jurkat cells were treated with 10 ?M CA-074Me for 3 hours (blue histogram), 10?M Leupeptin for 1 hour (red histogram) or were untreated (green histogram). They were then stained for 1 hour with R110-(RR)2. After incubation with the R110-(RR)2 substrate, the samples were analyzed by flow cytometry using a ZE5 Cell Analyzer (488 nm excitation and a 525/35 emission filter). Treatment with CA-074Me or Leupeptin decreased the fluorescence signal detected compared to the untreated control (see table), which displayed a baseline level of cathepsin activity)

(ICT's Caspase 8 FLICA kit was used todetect apoptosis in U937 cells, ahistiocytic lymphoma cell line. Cells weretreated with a control, 50 ng/mL TNF, or50 ug/mL cycloheximide (CHX) for 24hours. Cells were then incubated withFAM-LETD-FMK for 1 hour, washed 4times, then analyzed on a flowcytometer. (IN VIVO APOPTOSIS IN MOUSE BRAIN. Here are whole animal images using FLIVO to study brain infections. Image take with a Carestream In Vivo FX PRO imager. Adult wild-type Balb/c mice were either given a control (#1&2 top and middle) or intranasal inoculation with the HSV-1 virus (#3 bottom), which is absorbed thru the epithelium and is known to induce apoptosis in the brain. 7 days later, at the peak of infection, the mice were injected IV in the tail vein without any reagent (#1) or with NIR-690 (#2 and #3 middle and bottom), and imaged 4 hours later. We can see strong caspase activity located in the brain of the bottom animal treated with HSV-1 and injected with NIR690 (#3 bottom). Minimal signal was detected in the liver region of both mice injected with NIR690 (#2 and #3) as the liver is the route of clearance for FLIVO tracers. There is no signal in the uninfected control (#1 top), therefore #2 (middle) reveals background staining.)
