Background: Prostaglandin F2-alpha, One of the prostaglandins, a group of hormone-like substances that participate in a wide range of body functions such as the contraction and relaxation of smooth muscle, the dilation and constriction of blood vessels, control of blood pressure, and modulation of inflammation. Prostaglandin F2-alpha(PGF-2 alpha) is a stable prostaglandin that stimulates the contraction of uterine and bronchial smooth muscle and produces vasoconstriction (tightening) in some blood vessels.
Principle of the Assay: This kit was based on Competitive-ELISA detection method. The microtiter plate provided in this kit has been pre-coated with PGF2alpha. During the reaction, PGF2alpha in the sample or standard competes with a fixed amount of PGF2alpha on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to PGF2alpha. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzymesubstrate reaction is terminated by the addition of a acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of PGF2alpha in the samples is then determined by comparing the OD of the samples to the standard curve. The concentration of the target substance was inversely proportional to the OD450 value.
