ANKS6 is a protein containing nine N-terminal ankyrin repeats and a C-terminal sterile alpha motif (SAM). An Arg823Trp missense mutation of Anks6 (SamCystin, Pkdr1) has been identified as the underlying cause of cystic kidney disease in the Han:SPRD +/Cy rat. ANKS6 requires NEK8 for localizing to the ciliary inversin compartment (IC) and activates NEK8 by binding to its kinase domain. ANKS6 is downstream of NEK8 in the IC assembly hierarchy, and exhibits a uniquely strong and functional interaction with NEK8 that is necessary for a NEK8-mediated phosphorylation pathway.
