Proper function of GABAergic synapses depends upon the postsynaptic compartment anchoring of neurotransmitter receptors to the membrane by gephyrin and collybistin (Cb). In humans, collybistin is encoded by ARHGEF9 on Xq11.1. Collybistin is a guanine nucleotide exchange factor for Rho family GTPases that is crucial in gephyrin clustering and GABAA receptor localization. Collybistin can regulate actin cytoskeletal dynamics and may also modulate GABAergic and glycinergic neurotransmission through binding of a scaffolding protein, gephyrin, at the synapse. ARHGEF9 plays an essential role in the mammalian brain. ARHGEF9 mutations have been identified in patients with heterogeneous phenotypes, including epilepsy of variable severity and intellectual disability.
