Preparation and Storage
Store at -20 degree C for one year from date of receipt. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for six months. Avoid repeated freezing and thawing.

WB (Western Blot)

(Figure 1. Western blot analysis of CD9 using anti-CD9 antibody (AAA126124).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: human HCCT tissue lysates,Lane 2: human RT4 whole cell lysates.After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD9 antigen affinity purified polyclonal antibody (#AAA126124) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CD9 at approximately 22 kDa. The expected band size for CD9 is at 25 kDa.)

Principle of the Assay: The Picokine™ Human CA13 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Human CA13 with a 96-well strip plate that is pre-coated with antibody specific for CA13. The detection antibody is a biotinylated antibody specific for CA13. The capture antibody is monoclonal antibody from mouse and the detection antibody is biotinylated polyclonal antibody from goat. The kit contains recombinant Human CA13 with immunogen: Expression system for standard: E.coli; Immunogen sequence: M1-H262. The kit is analytically validated with ready-to-use reagents. To measure Human CA13, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Human CA13 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human CA13 in the sample.
