WB (Western Blot)
(Figure 1. Western blot analysis of Dynamin 2/DNM2 using anti-Dynamin 2/DNM2 antibody (AAA126158).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: human RT4 whole cell lysates,Lane 2: human MCF-7 whole cell lysates.After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Dynamin 2/DNM2 antigen affinity purified polyclonal antibody (#AAA126158) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Dynamin 2/DNM2 at approximately 100 kDa. The expected band size for Dynamin 2/DNM2 is at 98 kDa.)

Background: Neural Cell Adhesion Molecule, NCAM, also known as the cluster of differentiation CD56, is a hemophilic binding glycoprotein. It is a glycoprotein of Immunoglobulin(Ig) super family. At least 27 alternatively spliced NCAM mRNAs are produced, giving a wide diversity of NCAM isoforms NCAM gene is located at 11q22-q23. This glycoprotein is mainly expressed on the surface of neurons, glia, skeletal muscle and natural killer cells. NCAM has been implicated as having a role in cell-cell adhesion, neurite outgrowth, synaptic plasticity, and learning and memory.
Principle of the Assay: The Quick Picokine™ Human NCAM1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human NCAM1 in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). It uses our proprietary Quick ELISA technology. Quick ELISA technology employs capture antibodies conjugated to an affinity tag that is recognized by the polyclonal antibody used to coat our Quick ELISA plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The kit contains recombinant Human NCAM1 with immunogen: Expression system for standard: NS0; Immunogen sequence: L20-G718. To measure Human NCAM1, add standards and samples to the wells, then add antibody cocktail. Wash the wells with PBS or TBS buffer, and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product is linearly proportional to Human NCAM1 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human NCAM1 in the sample.
