IF (Immunofluorescence)
(AAA328899 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(AAA328899 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
IHC (Immunohiostchemistry)
(AAA328899 at 1/100 staining Mouse kidney tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)
WB (Western Blot)
(Western blot analysis of extracts from MCF-7, using LOX Antibody. The lane on the left was treated with blocking peptide.)
Post Translational Modifications: The lysine tyrosylquinone cross-link (LTQ) is generated by condensation of the epsilon-amino group of a lysine with a topaquinone produced by oxidation of tyrosine.Proteolytically cleaved by BMP1 which removes the propeptide (PubMed:31152061). Also proteolytically cleaved by ADAMTS2 and ADAMTS14, but not by ADAMTS3, at an additional cleavage site downstream of the BMP1 cleavage site (PubMed:31152061). The propeptide plays a role in directing the deposition of this enzyme to elastic fibers, via interaction with tropoelastin (By similarity). Cleavage by BMP1 to remove the propeptide does not increase enzymatic activity but increases binding to collagen (PubMed:31152061). Cleavage by ADAMTS2 produces a form with reduced collagen-binding activity (PubMed:31152061).Sulfated at Tyr-187 and also at either Tyr-183 or Tyr-184 which enhances binding to collagen.
Subcellular Location: Secreted. Secreted>Extracellular space.
Tissue Specificity: Heart, placenta, skeletal muscle, kidney, lung and pancreas.
Subunit Structure: Interacts with MFAP4 (PubMed:26601954).
Similarity: Belongs to the lysyl oxidase family.

Product Notes
The LOX lox (Catalog #AAA328899) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The LOX Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(88%), Horse(100%), Rabbit(100%), Dog(100%), Chicken(88%), Xenopus(88%) and may cross-react with other species as described in the data sheet. AAA Biotech's LOX can be used in a range of immunoassay formats including, but not limited to, ELISA, ICC (Immunocytochemistry), IF (Immunofluorescence). Researchers should empirically determine the suitability of the LOX lox for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "LOX, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
