ChIP (Chromatin Immunoprecipitation)

(NFkB p65 (pAb) tested by ChIP. ChIP was performed using the ChIP-IT High Sensitivity Kit with 2 million HT29 cells treated with TNF and 10 ul of p65 antibody. ChIP DNA was used in qPCR with the negative control primer pair or gene-specific primer pairs as indicated. Data are presented as Binding Events Detected per 1000 Cells using Epigenetic Services normalization scheme which accounts for primer efficiency and the amount of chromatin used in the ChIP reaction.)

Short Description: Our NFkB p65 antibody (pAb) was raised in a Rabbit host. It has been validated for use in Chromatin Immunoprecipitation, ChIP-Seq, Supershift and Western blot; it has been shown to react with Human samples.

Background: NFkB p65 (Nuclear factor NF-kappa-B p65 subunit also known as NFkB p65 or RelA) is a member of the Rel family of transcription factors that are involved in processes such as inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis. The Rel family includes c-Rel, RelA (NFkB p65, NFkB3) and RelB (I-Rel). Rel proteins form dimers with the NFkB p50 (proteolytically processed from p105/NFkB1) and p52 (proteolytically processed from p100/NFkB2) members of the NFkB family that bind DNA and activate transcription. The NFkB p65/p50 heterodimer is the most abundant form of NFkB. NFkB signaling is negatively regulated by the sequestration of the NFkB complex in the cytoplasm by its association with the IkB family of inhibitory proteins. The IkB Kinase (IKK) complex is the key enzyme involved in the activation and translocation of NFkB. The IKK complex is composed of two catalytic subunits (IKKa and IKKb) and a regulatory subunit (IKKg). Upon its activation, the IKK complex phosphorylates IkB proteins (IkBa, IkBa phospho Ser32, 36), which marks them for ubiquitination and degradation. This enables the NFkB complex to translocate to the nucleus where it regulates transcription by binding to DNA.

Product Notes

The NFkBp65 rela (Catalog #AAA59863) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The NFkB p65 antibody (pAb) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's NFkB p65 can be used in a range of immunoassay formats including, but not limited to, WB (Western Blot), EMSA (Electrophoretic Mobility Shift Assay), ChIP (Chromatin immunoprecipitation), ChIP (Chromatin immunoprecipitation). Researchers should empirically determine the suitability of the NFkBp65 rela for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NFkB p65, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
