Protein Function
Major glycoprotein of vascular endothelium. Involved in the regulation of angiogenesis. May play a critical role in the binding of endothelial cells to integrins and/or other RGD receptors. Acts as TGF-beta coreceptor and is involved in the TGF- beta/BMP signaling cascade. Required for GDF2/BMP9 signaling through SMAD1 in endothelial cells.
Preparation and Storage
At -20 degree C for one year from date of receipt. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for six months.Avoid repeated freezing and thawing.

WB (Western Blot)

Western blot analysis of Ninein/NIN using anti-Ninein/NIN antibody (AAA127154).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: human Hela whole cell lysates,Lane 2: human MOLT4 whole cell lysates,Lane 3: human MCF-7 whole cell lysates,Lane 4: human U20S whole cell lysates,Lane 5: rat heart tissue lysates,Lane 6: mouse heart tissue lysates.After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Ninein/NIN antigen affinity purified polyclonal antibody (#AAA127154) at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Ninein/NIN at approximately 260 kDa. The expected band size for Ninein/NIN is at 243 kDa.)
