Funtion
Cooperates with LY96 and CD14 to mediatethe innate immune response to bacteriallipopolysaccharide (LPS) (PubMed: ). Actsvia MYD88, TIRAP and TRAF6, leading toNF-kappa-B activation, cytokine secretionand the inflammatory response (PubMed:9237759,PubMed: ,PubMed: ). Alsoinvolved in LPS-independent inflammatoryresponses triggered by free fatty acids, such as palmitate, and Ni(2+). Responsestriggered by Ni(2+) require non- conservedhistidines and are, therefore,species-specific (PubMed: ). BothM.tuberculosis HSP70 (dnaK) and HSP65(groEL-2) act via this protein to stimulateNF-kappa-B expression (PubMed: ). Incomplex with TLR6, promotes sterileinflammation in monocytes/macrophages inresponse to oxidized low-density lipoprotein(oxLDL) or amyloid-beta 42. In this context, the initial signal is provided by oxLDL- oramyloid-beta 42-binding to CD36. Thisevent induces the formation of aheterodimer of TLR4 and TLR6, which israpidly internalized and triggersinflammatory response, leading to theNF-kappa-B-dependent production ofCXCL1, CXCL2 and CCL9 cytokines, viaMYD88 signaling pathway, and CCL5cytokine, via TICAM1 signaling pathway, aswell as IL1B secretion. Bindselectronegative LDL (LDL(-)) and mediatesthe cytokine release induced by LDL(-)(PubMed: ).Stimulation of monocytes in vitro withM.tuberculosis PstS1 induces p38 MAPK andERK1/2 activation primarily via TLR2, butalso partially via this receptor (PubMed: ).
Cellular Location
Cell membrane; Single-pass type Imembrane protein. Early endosome. Cellprojection, ruffle{ECO:0000250|UniProtKB:Q9QUK6}.Note=Upon complex formation with CD36and TLR6, internalized throughdynamin-dependent endocytosis(PubMed:20037584). Colocalizes withRFTN1 at cell membrane and then togetherwith RFTN1 moves to endosomes, uponlipopolysaccharide stimulation.
Tissue Location
Highly expressed in placenta, spleen andperipheral blood leukocytes(PubMed:9435236, PubMed:9237759).Detected in monocytes, macrophages, dendritic cells and several types of T-cells(PubMed:9237759, PubMed:27022195).
Preparation and Storage
Maintain refrigerated at 2-8°C for up to 2 weeks. For long term storage store at -20°C in small aliquots to prevent freeze-thaw cycles.

Related Product Information for anti-TLR4 antibody
TLR4 is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This receptor is most abundantly expressed in placenta, and in myelomonocytic subpopulation of the leukocytes. It has been implicated in signal transduction events induced by lipopolysaccharide (LPS) found in most gram-negative bacteria.

Product Notes

The TLR4 tlr4 (Catalog #AAA286138) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 669-698. The TLR4 Antibody (Center) reacts with Human Predicted Reactivity: Bovine, Hamster, Horse, Pig and may cross-react with other species as described in the data sheet. AAA Biotech's TLR4 can be used in a range of immunoassay formats including, but not limited to, FCM/FACS (Flow Cytometry), ELISA, WB (Western Blot). Researchers should empirically determine the suitability of the TLR4 tlr4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TLR4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
