Ubiquilin-2 (UBQLN2) is a 66?kDa protein member of the ubiquilin family. UBQLN2 has a ubiquitin-like domain (UBL) in its N-terminal that can interact with the proteasome, and a ubiquitin-associated domain (UBA) on the C-terminal which recognizes the polyubiquitin chains on marked proteins. UBQLN2 contains four stress-induced protein 1-like domains (STI-1 like), located between the residues 178-247 and 379-462, which are involved in the interaction with heat-shock proteins and autophagy mediators. UBQLN2 has also a unique PXX domain containing 12 tandem repeats implicated in protein-protein interactions. This domain differentiates UBQLN2 from other members of the ubiquilin family. UBQLN2 plays an essential role in noxious protein accumulation and clearance pathways in amyotrophic lateral sclerosis and frontotemporal dementia (ALS/FTD) pathogenesis.
