Specificity
This assay has high sensitivity and excellent specificity for detection of PGC1alpha. No significant cross-reactivity or interference between PGC1alpha and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between PGC1alpha and all the analogues, therefore, cross reaction may still exist in some cases.

Principle of the Assay: PGC1alpha ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-PGC1alpha antibody and an PGC1alpha-HRP conjugate. The assay sample and buffer are incubated together with PGC1alpha-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PGC1alpha concentration since PGC1alpha from samples and PGC1alpha-HRP conjugate compete for the anti-PGC1alpha antibody binding site. Since the number of sites is limited, as more sites are occupied by PGC1alpha from the sample, fewer sites are left to bind PGC1alpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PGC1alpha concentration in each sample is interpolated from this standard curve.

Product Notes
The Human PPARgC1a ppargc1a (Catalog #AAA247215) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA247215 ELISA Kit recognizes Human PPARgC1a. It is sometimes possible for the material contained within the vial of "Peroxisome Proliferator Activated Receptor Gamma Coactivator 1 Alpha, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
