Epidermal growth factor receptor (EGFR) and the adaptor protein Shc were reported to be substrates of this PTP, which suggested the roles in growth factor mediated cell signaling. PTPN1 acts as a negative regulator of insulin signaling by dephosphorylating the phosphotryosine residues of insulin receptor kinase. PTPN1 dephosphorylates epidermal growth factor receptor kinase, as well as JAK2 and TYK2 kinases.
