Principle of the Assay: Rat Catalase (CAT) ELISA Kit employs a two-site sandwich ELISA to quantitate Catalase in samples. An antibody specific for Catalase has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Catalase present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Rat Catalase detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of Catalase bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background/Introduction: Catalase is a common enzyme found in nearly all living organisms exposed to oxygen (such as bacteria, plants, and animals). It catalyzes the decomposition of hydrogen peroxide to water and oxygen. It is a very important enzyme in protecting the cell from oxidative damage by reactive oxygen species (ROS). Likewise, catalase has one of the highest turnover numbers of all enzymes; one catalase molecule can convert millions of hydrogen peroxide molecules to water and oxygen each second.
