Principle of the assay: Rat CD86 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for CD86 has been precoated onto 96-well plates. Standards(NSO, V29-I250) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CD86 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the rat CD86 amount of sample captured in plate.
Background: Cluster of Differentiation 86 (also known as CD86 and B7-2) is aprotein expressed on antigen-presenting cells that provides costimulatory signals necessary for T cell activation and survival. The CD86 gene encodes a type I membrane protein that is a member of the immunoglobulin superfamily. Using fluorescence in situ hybridization mapping, the CD86, like CD80, was mapped to human 3q21 and mouse chromosome 16, band B5. The antigen presentation coactivators B71 and B72, which are important in other immune-mediated thyroid diseases, are important for lymphocytic infiltration and the immune response against thyroid carcinoma.
