Principle of the Assay: Interleukin-1 (IL-1) is one of the first cytokines ever described. Its initial discovery was as a factor that could induce fever, control lymphocytes, increase the number of bone marrow cells and cause degeneration of bone joints. At this time, IL-1 was knownunder several other names including endogenous pyrogen, lymphocyte activating factor, haemopoetin-1 and mononuclear cell factor, amongst others.Rat IL-1beta ELISA Kit employs a double antibody sandwich method to quantitate Rat IL-1beta insamples. An antibody specific for Rat IL-1beta has been pre-coated onto a microplate. Standards and samples are pipetted into thewells and any Rat IL-1beta present is bound by the immobilized antibody. After removing any unbound substances, abiotin-conjugated antibody specific for Rat IL-1beta is added to the wells. After washing, proprietary Streptavidin-HRP conjugates isadded to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMBturns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with a microplate readerat 450 nm wavelength. The IL-1beta concentration is proportional to the OD450 nm value.
