Background: Serotonin (5-hydroxytryptamine, 5-HT) is a central nervous system monoamine implicated in a number of physiological functions, including sleep, mood, appetite, anxiety, and neurovegetative control.Serotonin acts through the activation of G protein receptors called serotonin receptors (5-HTRs).Seven 5-HTR subclasses (5-HTR1-7) have been described to date.Serotonin receptor 4 (5-HTR4) couples to Galphas(3),with stimulation of the receptor leading to adenylyl cyclase activation, elevation of cyclic AMP levels and downstream PKA activation.The corresponding HTR4 gene exhibits a complex structure, which includes a large number of splice variants that seem to share the same pharmacological properties. 5-HTR4 is one of several receptors implicated in cognition, depression,and the development of Alzheimer's disease.
Principle of the Assay: This kit was based on Competitive-ELISA detection method. The microtiter plate provided in this kit has been pre-coated with ST/5-HT. During the reaction, ST/5-HT in the sample or standard competes with a fixed amount of ST/5-HT on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to ST/5-HT. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of ST/5-HT in the samples is then determined by comparing the OD of the samples to the standard curve. The concentration of the target substance was inversely proportional to the OD450 value.
