Components
Safranin staining solution: 100 ml
Fast green staining solution: 100 ml

Protocol
1. Dewax the tissue section in dimethylbenzene for 10 min, then, immerse the section for another 10 min in the fresh dimethylbenzene.
2. Immerse the tissue section in 100%, 95% & 75% ethanol for 1 min respectively, then, rinse the section with water.
3. Add Fast green staining solution to the section for about 15-20 min (Ossification is dark green, meanwhile, the cartilage is colorless), wash slightly by water. (DO NOT wash section with long time, since the fast green would be faded easily).
4. Add Safranin staining solution to the section for about 1-3 min until cartilage turns bright red.
5. Wash the redundant staining solution slightly by water.
6. Immerse the section into Color separation solution (Anhydrous ethanol, Prepared by user) for 10s-10min. (Please observe the section carefully in this process to control the staining degrees of these 2 colors).
7. Dry the section and seal with neutral gum.

Staining results
Cartilage turns bright red, ossification turns green. Some connective tissues are red too.
Note
1. If the Safranin staining solution make the section in dark red and covers the green, it is recommended to elute the red with 1% hydrochloric acid alcohol, then stain again.
2. If the fast green got the light staining, end users can stain the section again with Fast gree staining solution after Step 6.
3. Pay attention to control the staining degree of the 2 colors under a microscope, since the slight staining time differences would make big differences between red and green.
4. If the Safranin staining solution was used repeatedly for many circles, it may got sediment which leads to the difficult staining. In this way, please replace the solution to ensure the good results.
