Background: 2'-5'-oligoadenylate synthetase 1 (OAS1) is an antiviral protein induced by type 1 interferon that plays a key role in the cellular innate immune response. The OAS1 enzyme produces the second messenger 2'-5'-linked oligoadenylate in response to cytosolic dsRNA. These 2'-5'-linked oligoadenylates bind to the ribonuclease RNase L, which then degrades viral and cellular RNA. Polymorphisms in the corresponding OAS1 gene have been examined for possible association with increased susceptibility to type 1 diabetes mellitus, multiple sclerosis, and infection by viral pathogens.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti 2, 5-OAS antibody was pre-coated onto the 96-well plate. The biotin conjugated anti 2, 5-OAS antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with 2, 5-OAS conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of 2, 5-OAS in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
