Background: Angiopoietin is a secreted glycoprotein. There are now four identified angiopoietins: ANGPT1, ANGPT2, ANGPTL3, ANGPT4. Angiogenesis proceeds through sprouting, endothelial cell migration,proliferation, and vessel destabilization and stabilization. They are responsible for assembling and disassembling the endothelial lining of blood vessels. Angiopoietin cytokines are involved with controlling microvascular permeability, vasodilation, and vasoconstriction by signaling smooth muscle cells surrounding vessels.

Principle of the Assay: ANG2 ELISA kit applies the solid phase sandwich enzyme immunoassay technique. The microplate has been pre-coated with antibody specific to ANG2. Standards or samples are then added to the microplate wells and ANG2 if present, will bind to the antibody pre-coated wells. Then a biotin-conjugateddetection antibody specific toANG2 is added to the wells to form a pre-coatedantibody-antigen-detection antibody Complex. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells, and then a TMB substrate solution is added to the wells to incubate. TMB substrate solution turns to blue after oxidating by HRP, and finally turns to yellow after adding the stop solution. Color develops in proportion to the amount of ANG2 bound in the initial steps. The color is measured spectrophotometrically at a wavelength of 450nm. The concentration of ANG2 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
