Principle & Procedure
plasmid purfication protocols are based on a modified alkaline lysis procedure, followed by binding of plasmid DNA to resin under appropriate low-salt and pH conditions. RNA, proteins, dyes, and low-molecular-weight impurities are removed by a medium-salt wash. Plasmid DNA is eluted in a high-salt buffer and then concentrated and desalted by isopropanol precipitation.
Determation of Yield
To determine the yield, DNA concentration should be determined by both UV spectrophotometry at 260 nm and quantitative analysis on an agarose gel. For reliable spectrophotometric DNA quantification, A260 readings should lie between 0.1 and 1.0
Note
Agarose gel analysis-We recommend removing and saving aliquots during the purification procedure. If the plasmid DNA is of low yield or quality, the samples can be analyzed by agarose gel electrophoresis to determine the stage of the purification procedure where the problem occurred.
Preparation and Storage
Spin Column CM should be stored dry and at room temperature (15-25 degree C). They can be stored for at least 2 years without showing any reduction in performance, capacity, or quality of separation. Fine Plasmid Kits should be stored at room temperature. After adding RNase A, Buffer P1 should be stored at 2-8 degree C and is stable for 6months. Other buffers and RNase A stock solution can be stored for 2 years at room temperature.
Related Product Information for BCA assay kit
Background: The BCA protein assay is a protein determination formulation based on bicinchoninic acid (BCA) for the colorimetric detection. This method combines the reduction of Cu2+ to Cu+ by protein in an alkaline medium (the biuret reaction) and the soluble purple-colored reaction product from the complexing of Cu+ and BCA. This purple-colored complex exhibits a maximum absorbance at 562 nm that is nearly linear with increasing protein concentrations over a broad working range (0-2 mg/ml). According to highly sensitivity and simple to use, the BCA protein assay is adopted by laboratories and companies, and becomes one of the prior protein quantitative methods as well as the Bradford assay. The BCA Protein Assay Kit has the characteristics of high sensitivity and light background, and the measured range can be up to 3 mg/ml.

Product Notes
The BCA (Catalog #AAA249416) is an Assay Kit and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "BCA Protein, Assay Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
