In vitro
Apical-to-basolateral exchange across endothelial monolayer of fluorescent Abeta42 (FAM-labeled Human beta-Amyloid (1-42) and Fluor 488-labeled beta-Amyloid (1-42)) or scramble Abeta42 (FAM-labeled scrambled beta-Amyloid (1-42))(1-100 uM) is monitored in transendothelial electrical resistance (TEER) during cell monolayer's formation over time (over 120 min in presence or absence of Abeta24). Human beta-Amyloid (1-24)(1 uM) results in H-Abeta42 retention, thus reducing its efflux through the BBB and therefore preventing an efficient mechanism of Abeta42 clearance[1].The in vitro BBB model is used to investigate the passage of FAM-labeled or 488-conjugated H-Abeta42 across the endothelial cell monolayer. The addition of fluorescently labeled H-Abeta42 (FAM-labeled Human beta-Amyloid (1-42) or Fluor 488-labeled beta-Amyloid (1-42)) to the apical side of cell inserts results in effective BBB crossing, FAM-labeled scrambled beta-Amyloid (1-42) is not efficiently transcytosed.

Product Notes
The beta-Amyloid (Catalog #AAA227857) is a Biochemical and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "beta-Amyloid, Biochemical" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
