Principle of the Assay: This kit was based on Capture-ELISA detection method. Anti Canine IgM (u chain) was pre-coated onto the 96-well plate. The biotin conjugated antigen was used as the detection antigen. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotin detection antigen was added to bind with Anti-KLH IgM conjugated on Anti Canine IgM (u chain).Excess conjugate and unbound sample or standard are washed from the plate, and HRPStreptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of Anti-KLH IgM in the samples is then determined by comparing the OD of the samples to the standard curve. The concentration of the target substance was inversely proportional to the OD450 value.
