Background: Chikunguya Virus (CHIKV) is a member of the Alphavirus genus. It is also classified as an Arbovirus due to vector-dependent t ransmission by mosquitos. The enveloped v irus carries structural glycoproteins E1-E3 which are immunogenic. S everal non-structural proteins (nsP) and a ss-(+) RNA genome are present within the viral capsid. Chikungunya f ever i s c haracterized by f ever, joint pai n an d swelling, r ash, an d f lu-like symptoms. Symptoms usually appear within a period of 1-2 weeks post-infection. The disease is characterized by a biphasic course with an initial stage of high virus load and a convalescent stage without viremia. I n rare cases, neurological d isorders like Guillain-Barré syndrome and meningoencephalitis have occurred.
Principle of the Assay: The ELISA (Enzyme-Linked Immunosorbent Assay) is an immunoassay suited to the detection of ant ibodies. T he r eaction i s bas ed on t he specific i nteraction of antibodies w ith t heir corresponding antigen. The test strips of the SERION ELISA classic microtiter plate are coated with specific antigens of the pathogen of interest. If antibody in a sample is present, they bind to the fixed antigen. A secondary antibody, which has been conjugated with the enzyme alkaline phosphatase, detects and binds to the antigen-antibody c omplex. T he c olorless s ubstrate p - nitrophenylphosphate i s t hen c onverted i nto t he c olored pr oduct p -nitrophenol. T he s ignal intensity of t his reaction pr oduct i s pr oportional t o the c oncentration of t he an tibody i n t he sample and is measured photometrically.
