product-image-AAA152273_BIOACTIVITY13.jpg Bioactivity (Brain-derived neurotrophic factor, also known as BDNF, is a member of the neurotrophin family of growth factors, which are related to the canonical Nerve Growth Factor. BDNF acts on certain neurons of the central nervous system and the peripheral nervous system, helping to support the survival of existing neurons, and encourage the growth and differentiation of new neurons and synapses. Besides, Sortilin 1 (SORT1) has been identified as an interactor of BDNF, thus a binding ELISA assay was conducted to detect the interaction of recombinant human BDNF and recombinant human SORT1. Briefly, SORT1 were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100ul were then transferred to BDNF-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-SORT1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450/630nm immediately. The binding activity of of BDNF and SORT1 was shown in Figure 1, and this effect was in a dose dependent manner.)

Product Notes

The BDNF bdnf (Catalog #AAA152273) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's Brain Derived Neurotrophic Factor (BDNF) can be used in a range of immunoassay formats including, but not limited to, Activity Assay, Cell Culture. Researchers should empirically determine the suitability of the BDNF bdnf for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: His129~Arg 247. It is sometimes possible for the material contained within the vial of "Brain Derived Neurotrophic Factor (BDNF), Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
