• Materials Provided: 1. Capture Antibody 1 mg/mL of rabbit anti-VSIG4 monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 2ug/mL in PBS before coating. ; 2. Detection Antibody 0.25 mg/mL rabbit anti-VSIG4 monoclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-protector, pH 7.4). Dilute to working concentration of 0.25ug/mL in detection antibody dilution buffer before use. ; 3. Standard Each vial contains 100 ng of recombinant VSIG4. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20 degree C to -80 degree C in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 2ng/mL is recommended.

Background: VSIG4 (V-set and immunoglobulin domain containing 4), also known as complement receptor of the immunoglobulin superfamily (CRIg) and Z39Ig, is a type I transmembrane glycoprotein. It is a B7 familyrelated protein and an Ig superfamily member. In contrast to the B7 family members which contain two IgG domains, VSIG4 contains one complete V-type I g domain and a truncated C-type I g domain. VSIG4 is exclusively expressed on tissue resident macrophages and binds to multimers of C3b and iC3b that are covalently attached to particle surfaces. No VSIG4 expression appears to be present in T and B cells. VSIG4 functions as a negative regulator of T cell activation, and may be involved in the maintenance of peripheral T cell tolerance, and is also identified as a potent suppressor of established inflammation. Mouse VSIG4 is synthesized as a 28 amino acid precursor that contains a signal sequence, an V-type I g domain (aa 36-115), one potential N-linked glycosylation site, and a single transmembrane domain. The V-type I g domain of mouse VSIG4 shares 86% and 8% aa sequence identity with the V-type I g domains of rat and human VSIG4, respectively.

Principle of the Assay: The ELISA Pair Set is a solid phase sandwich ELISA (Enzyme-Linked Immunosorbent Assay). It utilizes a monoclonal antibody specific for Human VSIG4 coated on a 96-well plate. Standards and samples are added to the wells, and any Human VSIG4 present binds to the immobilized antibody. The wells are washed and a horseradish peroxidase conjugated rabbit anti-Human VSIG4 monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of Human VSIG4 present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.

Intended Uses: The Human VSIG4 ELISA Pair Set is for the quantitative determination of Human VSIG4. This ELISA Pair Set contains the basic components required for the development of sandwich ELISAs.
