Background: Vitamin D-binding protein (DBP), also/originally known as gc-globulin (group-specific component), is a protein that in humans is encoded by the GC gene. It is mapped to 5 E1; 5 44.32 cM. The protein encoded by this gene belongs to the albumin gene family. It is a multifunctional protein found in plasma, ascitic fluid, cerebrospinal fluid and on the surface of many cell types. It binds to vitamin D and its plasma metabolites and transports them to target tissues. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.

Principle of the Assay: The Picokine™ Mouse Gc Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Mouse Gc with a 96-well strip plate that is pre-coated with antibody specific for Gc. The detection antibody is a biotinylated antibody specific for Gc. The capture antibody is a monoclonal antibody from rat and the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Mouse Gc with immunogen: Expression system for standard: NS0; Immunogen sequence: L17-S476. The kit is analytically validated with ready-to-use reagents. To measure Mouse Gc, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Mouse Gc in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Gc in the sample.
