Purity/Purification
>92% as determined by SDS-PAGE
Form/Format
Lyophilized from sterile PBS, pH7.4. Normally 5%-8% trehalose, mannitol and 0.01% Tween80 are added as protectants before lyophilization. Please refer to the specific buffer information in the hard copy of CoA.
Sequence
Met1-Thr211
Species
Human
Activity
1. Measured by its binding ability in a functional ELISA. Immobilized human TNFRI-His at 10ug/mL (100uL/well) can bind biotinylated human TNFalpha. The EC50 of biotinylated human TNFalpha is 5-12ug/mL.
2. Measured by its ability to inhibit TNFalpha-mediated cytotoxicity in L929 mouse fibrosarcoma cells in the presence of metabolic inhibitor actinomycin D. The ED50 for this effect is typically 0.2-1ug/mL in the presence of 0.25ng/mL recombinant human TNFalpha.
Endotoxin
<1.0EU per ug of the protein as determined by the LAL method
Predicted N Terminal
Ile 22
Tag
C-His
Protein Construction
A DNA sequence encoding the human TNFRI (NP_001056.1) extracellular domain (Met 1-Thr 211) was fused with a polyhistidine tag at the C-terminus.
Reconstitution
A hardcopy of COA with reconstitution instruction is sent along with the products. Please refer to it for detailed information.
Preparation and Storage
Samples are stable for up to twelve months from date of receipt at -20 degree C to -80 degree C. Store it under sterile conditions at -20 degree C to -80 degree C. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.
In general, recombinant proteins are provided as lyophilized powder which are shipped at ambient temperature. Bulk packages of recombinant proteins are provided as frozen liquid.
They are shipped out with blue ice.

Background: The cluster of differentiation (CD) system is commonly used as cell markers in Immunophenotyping. Different kinds of cells in the immune system can be identified through the surface CD molecules which associating with the immune function of the cell. There are more than 320 CD unique clusters and subclusters have been identified. Some of the CD molecules serve as receptors or ligands important to the cell through initiating a signal cascade which then alter the behavior of the cell. Some CD proteins do not take part in cell signal process but have other functions such as cell adhesion. CD120a (cluste of differentiation 120a), also known as TNFR1/TNFRSF1A, is a member of CD family, tumor necrosis factor receptor superfamily. CD120a is one of the most primary receptors for the tumor necrosis factor-alpha. It has been shown to be localized to both plasma membrane lipid rafts and the trans golgi complex with the help of the death domain (DD). CD120a can activate the transcription factor NF-kappaB, mediate apoptosis, and regulate inflammation processes.

Product Notes

The TNFRSF1A tnfrsf1a (Catalog #AAA257940) is an Active Protein produced from HEK293 Cells and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: Met1-Thr21 1. It is sometimes possible for the material contained within the vial of "TNFRSF1A, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
