Principle of the Assay: The LumiAb Human sCD40 Ligand Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human sCD40 Ligand concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human sCD40 Ligand while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human sCD40 Ligand, also known as Tumor Necrosis Factor Ligand Superfamily Member 5, is a 149 amino acid cytokine protein encoded by the CD40LG gene located at locus Xq26 on the X chromosome. The soluble CD40 ligand is generated when the membrane form of the CD40 ligand undergoes proteolytic processing to remove the first 112 residues from the N-terminal region. The homotrimeric soluble CD40 ligand Mediates B-cell proliferation in the absence of co-stimulus as well as IgE production in the presence of IL-4; it is involved in immunoglobulin class switching. Moreover, release of soluble CD40L from platelets is partially regulated by GP IIb/IIIa, actin polymerization, and the matrix metalloproteinases (MMP) inhibitor-sensitive pathway. Defects in CD40LG are the cause of X-linked immunodeficiency with hyper-IgM type 1 (HIGM1), also known as X-linked hyper IgM syndrome (XHIM). HIGM1 is an immunoglobulin isoform switch defect characterized by elevated concentrations of serum IgM and decreased amounts of all other isoforms. Affected males present at an early age (usually within the first year of life) recurrent bacterial and opportunistic infections, including Pneumocystis carinii pneumonia and intractable diarrhea due to cryptosporidium infection.

Product Notes
The Human TACI tnfrsf13b (Catalog #AAA318560) is a CLIA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA318560 CLIA Kit recognizes Human TACI. It is sometimes possible for the material contained within the vial of "TACI, CLIA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
