Principle of the Assay: beta-NGF is a member of the NGF-beta family and encodes a secreted protein which homodimerizes and is incorporated into a larger complex. This protein has nerve growth stimulating activity and the complex is involved in the regulation of growth and the differentiation of sympathetic and certain sensory neurons.Human beta-NGF ELISA Kit employs a double antibody sandwich method to quantitate Human beta-NGF in samples. An antibody specific for Human beta-NGF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Human beta-NGF present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Human beta-NGF is added to the wells. After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The beta-NGF concentration is proportional to the OD450 nm value.
