Principle of the Assay: The protein encoded by IL-1alpha gene is a member of the interleukin 1 cytokine family. This cytokine is a pleiotropic cytokine involved in various immune responses, inflammatory processes, and hematopoiesis.Human IL-1alpha ELISA Kit employs a double antibody sandwich method to quantitate Human IL-1alpha in samples. An antibody specific for Human IL-1alpha has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Human IL-1alpha present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Human IL-1alpha is added to the wells. After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450nm wavelength. The IL-1alpha concentration is proportional to the OD450 nm value.
