Background: Matrix metalloproteinase-14 is an enzyme that in humans is encoded by the MMP14 gene. Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Deficits in MMP14 leads to premature aging, short lifespan, and cell senescence in mice, suggesting an important role of MMP14 in extracellular matrix remodeling during aging.
Principle of the Assay: Human Matrix Metalloproteinase 14 (MMP-14) ELISA Kit employs a two-site sandwich ELISA to quantitate MMP-14 in samples. An antibody specific for MMP-14 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP-14 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human MMP-14 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of MMP-14 bound in the initial step. The color development is stopped and the intensity of the color is measured.
