Background: In 1957, American scientist Margoshoes isolated the metal protein of cadmium from animal organs while studying the role of metal biology. Because it is a low molecular weight, high sulfhydryl content and can bind a large amount of heavy metal ions, it is called metallothionein (referred to as MT). The MT molecule is oval in shape, divided into two domains, with a molecular weight of 6,000-7,000 Daltons, a diameter of 30-50Å, and contains 61 amino acids, of which 20 amino acids are cysteine, and a few have 21, so that each MT The molecule can bind 7 to 12 metal ions. Humans, animals, plants and microorganisms all contain MT, and their physical and chemical properties are basically the same, with special light absorption. The MT conformation is stronger and has strong heat resistance.
Principle of the Assay: Human Metallothionein-1G (MT1G) ELISA Kit employs a two-site sandwich ELISA to quantitate MT1G in samples. An antibody specific for MT1G has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MT1G present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human MT1G detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of MT1G bound in the initial step. The color development is stopped and the intensity of the color is measured.
