Background: The Microtubule-Associated Serine/Threonine Kinase family (MAST1-4, and MAST-like) is characterised by the presence of a serine/threonine kinase domain and a postsynaptic density protein-95/discs large/zona occludens-1 domain (PDZ). This latter domain gives the MAST family the capacity to scaffold its own kinase activity.
Principle of the Assay: Human Microtubule-associated serine/threonine-protein kinase 4 (MAST4) ELISA Kit employs a two-site sandwich ELISA to quantitate MAST4 in samples. An antibody specific for MAST4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MAST4 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human MAST4 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of MAST4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
