Background: NEDD8-activating enzyme E1 regulatory subunit encoded by NAE1 binds to the beta-amyloid precursor protein. Beta-amyloid precursor protein is a cell surface protein with signal-transducing properties, and it is thought to play a role in the pathogenesis of Alzheimer's disease. In addition, the encoded protein can form a heterodimer with UBE1C and bind and activate NEDD8, a ubiquitin-like protein. This protein is required for cell cycle progression through the S/M checkpoint. Three transcript variants encoding different isoforms have been found for NAE1.
Principle of the Assay: Human NEDD8-Activating Enzyme E1 Regulatory Subunit (NAE1) ELISA Kit employs a two-site sandwich ELISA to quantitate NAE1 in samples. An antibody specific for NAE1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NAE1 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human NAE1 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of NAE1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
