Recombinant human NAE1 (2-end) and full-length human UBA3 were expressed by baculovirus in Sf9 insect cells. NAE1 used an N-terminal His tag, and the gene accession number is NM_003905. UBA3 used an N-terminal GST tag, and the gene accession number is BC022853.
NAE1/UBA3 is a heterodimeric E1 (Ub-activating)-like complex that activates NEDD8. In the presence of ATP, NAE1/UBA3 adenylates and activates NEDD8 yielding NEDD8-UBA3 thioester and free AMP. Activated NEDD8 is then transferred to UBE2M or UBE2F through transthiolyation reaction. Finally, substrate specific-E3 ligase, such as RBX1/2 or DCN1, transfers NEDD8 from UBE2M or UBE2F to a lysine residue in its target protein (1,2). NEDD8 pathway plays an important role in cell cycle progression and survival (3).
