Background: PTP4A1 is highly expressed in fibroblasts from patients with SSc. PTP4A1 and its close homolog PTP4A2 are critical promoters of TGFbeta signaling in primary dermal fibroblasts and of bleomycin-induced fibrosis in vivo. PTP4A1 promotes TGFbeta signaling in human fibroblasts through enhancement of ERK activity, which stimulates SMAD3 expression and nuclear translocation.
Principle of the Assay: Human Protein tyrosine phosphatase type IVA 1 (PTP4A1) ELISA Kit employs a two-site sandwich ELISA to quantitate PTP4A1 in samples. An antibody specific for PTP4A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PTP4A1 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human PTP4A1 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of PTP4A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
