Background: Sdf-1, also known as chemokine CXCL12, is a small molecule of cytokine, belonging to the chemokine protein family. Chemokines have four conserved cysteine residues that form two pairs of disulfide bonds to form the specific structure of chemokines.The first and second cysteine residues are separated by an intervening amino acid residue.Sdf-1 comes in two forms, SDF-1alpha/CXCL12/CXCL12a and SDF-1alpha/CXCL12/CXCL12b.Sdf-1 alpha/CXCL12 receptor CXCR4 is a g-protein-coupled receptor with a seventh-order transmembrane structure.Sdf-1 alpha/CXCL12 binds to CXCR4 to stimulate receptor dimer formation and activate downstream signal transduction molecules, including ligate-adhesion kinase, extracellular signal-regulated kinase, protein kinase C, JAK/Stat, and NF-KB transcription pathways. Sdf-1 alpha /CXCL12/CXCR4 system has strong chemotaxis and activation effect on neutrophils, lymphocytes and eosinophils.This system plays an important role in regulating stem cell migration, cancer development, nervous system and cardiac repair.
Principle of the Assay: ELISA kit uses double antibody sandwich method: the anti-human SDF-1alpha/CXCL12 monoclonal antibody is coated on an enzyme plate. Human SDF-1alpha/CXCL12 monoclonal antibody is bound to human SDF-1alpha/CXCL12 monoclonal antibody in specimens and standard materials, and the free components are washed away. Biotinylated anti-HUMAN SDF-1alpha/CXCL12 antibody and horseradish peroxidase labeled avidin were added, and biotin specifically bound to avidin.The anti-human SDF-1alpha/CXCL12 antibody binds to human SDF-1alpha/CXCL12 to form an immune complex in which the free components are washed away. When a chromogenic substrate is added, if sdF-1 alpha/CXCL12 is present in the reaction well, horseradish peroxidase will make the colorless chromogenic agent appear blue, and the termination solution turn yellow.The concentration of SDF-1alpha/CXCL12 in human samples was positively proportional to OD450 when OD value was measured at 450 nm. The concentration of SDF-1alpha/CXCL12 in human samples could be calculated by drawing a standard curve.
