Background: Sphingosine kinase 1 is an enzyme that in humans is encoded by the SPHK1 gene. The protein encoded by this gene catalyzes the phosphorylation of sphingosine to form sphingosine-1- phosphate (S1P), a lipid mediator with both intra- and extracellular functions. Intracellularly, S1P regulates proliferation and survival, and extracellularly, it is a ligand for cell surface G protein-coupled receptors. This protein, and its product S1P, play a key role in TNF-alpha signaling and the NF-kappa-B activation pathway important in inflammatory, antiapoptotic, and immune processes. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.
Principle of the Assay: The Human SPHK1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human SPHK1 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human SPHK1 coated on a 96-well plate. Standards and samples are pipetted into the wells and SPHK1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human SPHK1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of SPHK1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Product Notes
The Human SPHK1 sphk1 (Catalog #AAA263086) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA263086 ELISA Kit recognizes Human SPHK1. It is sometimes possible for the material contained within the vial of "SPHK1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
