Principle of the Assay: TIMP metallopeptidase inhibit or 1, a member of the TIMP family. The glycoprotein is a natural inhibitor of the matrix metalloproteinases (MMPs), a group of peptidases involved in degradation of the extracellular matrix. In addition to its inhibitory role against most of the known MMPs.Human TIMP-1 ELISA Kit employs a double antibody sandwich method to quantitate Human TIMP-1 in samples. An antibody specific for Human TIMP-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Human TIMP-1 present is bound by the immobilized antibody. After removing any unbound substances, a HRP-conjugated antibody specific for Human TIMP-1 is added to the wells. After washing, remove any unbound HRP-conjugated antibody reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The TIMP-1 concentration is proportional to the OD450 nm value.
