Background: IFNgamma, also known as Interferon gamma or type II interferon, is a cytokine primarily produced by activated T cells and natural killer cells. It can stimulate various immune cells, such as macrophages, B cells, and NK cells, and has important immunoregulatory functions. IFNgamma can not only induce cellular antiviral responses, but also promote inflammatory reactions, enhance MHC molecule expression, induce apoptosis of tumor cells, and promote class switching of antibodies. IFNgamma plays an important role in immune responses. It can regulate the proliferation, differentiation, survival, and function of immune cells by activating various signaling pathways, and is closely related to the occurrence and development of various autoimmune diseases, infectious diseases, tumors, and other diseases.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti IFN-gamma antibody was precoated onto the 96-well plate. The biotin conjugated anti IFN-gamma antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with IFN-gamma conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of IFN-gamma in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
