Background: Interleukin 1beta (interleukin 1beta, IL-1beta) is a member of the interleukin 1 cytokine family, which is mainly secreted by activated monocytes, macrophages and epithelial cells. The generating precursor protein is hydrolyzed to its active form by caspase 1 (CASP1/ICE). As an important mediator of inflammatory response, it is involved in a variety of cellular activities, including cell proliferation, differentiation and apoptosis. IL-1betasignaling includes the binding of IL-1betato IL-1's auxiliary protein (IL-1-ACP), and then the complex binds to IL-1RI to activate the MAP kinase and NF-kappaB pathways.

Principle of the Assay: IL-1beta ELISA kit applies the solid phase sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with an antibody specific to IL-1beta. Standards or samples are then added to the microtiter plate wells and IL-1beta if present, will bind to the antibody pre-coatedwells.Then a biotin-conjugateddetection antibody specific to IL-1betais added to the wells to form a pre-coatedantibody-antigen-detection antibody Complex. After washing, Streptavidinconjugated Horseradish Peroxidase (HRP) is added to the wells, and then a TMB substrate solution is added to the wells to incubate. TMB substrate solution turns to blue after oxidating by HRP, and finally turns to yellow after adding the stop solution. Color develops in proportion to the amount of IL-1betabound in theinitial steps. The color is measured spectrophotometrically at a wavelength of 450nm. The concentration of IL-1betain the samples is then determined by comparing the O.D. of the samples to the standard curve.
