The Lymphocyte Antigen 6 Family Member G6D (LY6G6D) is a member of the LY6 family of proteins (typically 70-80 amino acids), mostly GPI-anchored to the cell surface. Regulated by the JAK-STAT5 and RAS-MEK-ERK cascades and belonging to a cluster of leukocyte antigens located in the MHC III region on chromosome 6, LY6G6D demonstrates selective expression in colorectal cancer (CRC), particularly the microsatellite stable subtype, with minimal to no expression in normal tissues. LY6G6D expression is also enhanced in diseases characterized by systemic immune suppression, such as autosomic dominant monocytopenia. LY6G6D up-regulation was predominant in CRCs characterized by an enrichment of immune suppressive regulatory T-cells and limited PD-1/PD-L1 immune checkpoint receptors, demonstrating its potential as a target for novel T-cell-dependent bispecific antibodies that can redirect T-cell effector function to colorectal tumors.
