Background: Neprilysin, also known as membrane metallo-endopeptidase, neutral endopeptidase (NEP), CD10, and common acute lymphoblastic leukemia antigen (CALLA), is a zinc-dependent metalloprotease enzyme that degrades a number of small secreted peptides, most notably theamyloid beta peptide whose abnormal misfolding and aggregation in neural tissue has been implicated as a cause of Alzheimer's disease, this gene is localized to human chromosome 3 by study of somatic cell hybrids and regionalized the location to 3q21-q27 by in situ hybridization. By cDNA transfection analysis, Neprilysin is confirmed as a functional neutral endopeptidase of the type that has previously been called enkephalinase. Neprilysin has also been called atriopeptidase. Atriopeptidase specifically degrades atrial natriuretic factor. A specific enzyme inhibitor was developed and reported that it had effects similar to those of low-dose ANF infusion. These effects include diuresis, natriuresis, vasodilatation, and suppression of the eninangiotensin-aldosterone system.
Principle of the Assay: The Picokine™ Mouse Mme Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Mouse Mme with a 96-well strip plate that is pre-coated with antibody specific for Mme. The detection antibody is a biotinylated antibody specific for Mme. The capture antibody is monoclonal antibody from rat and the detection antibody is polyclonal antibody from goat. The kit contains recombinant Mouse Mme with immunogen: Expression system for standard: NS0; Immunogen sequence: Y52-W750. The kit is analytically validated with ready-to-use reagents. To measure Mouse Mme, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Mouse Mme in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Mme in the sample.
