• Specificity: B220 isoform of CD45, Mr 240 kDa; CD45R/B220, a member of the protein tyrosine phosphate family and a major cell surface glycoprotein, represents a restricted form of the CD45 family which primarily recognizes only cells of B-lineage from pro-B cell through mature B lymphocytes and, prior to the availability of anti-CD19 MAbs, was commonly used as a pan B-cell marker.^1-5 It also reacts with certain activated T cells, as well as non-MHC-restricted lytically active lymphokine-activated killer (LAK) cells.^1, 6 In vivo administration of RA3-6B2 has been shown to affect differentiation of both T and B cells in normal mice and reduce the level of anti-DNA antibodies and lymphadenopathies in MRL/lpr mice.^7, 8

• Characterization: To ensure lot-to-lot consistency, each batch of monoclonal antibody is tested by flow cytometry to conform to characteristics of a standard reference reagent. Representative data are included in this product insert.
• Warning: Reagents contain sodium azide. Sodium is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
• Preparation and Storage: The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C. ; The fluorescein (FITC) conjugate is supplied as 0.5 mg or 0.1 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C. ; The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C. ; The R-phycoerythrin (R-PE) conjugate is supplied as 0.1 mg in 1.0 mL or 0.2 mg in 2.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze! ; The allophycocyanin (APC) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze! ; The Spectral Red (SPRD), R-phycoerythrin -Texas Red (R-PE-TXRD), R-phycoerythrin-Cyanine 7 (R-PE-CY 7), R-phycoerythrin-Cyanine 5.5 (R-PE-CY 5.5), Allophycocyanin-Cyanine 5.5 (APC-CY 5.5) and Allophycocyanin- Cyanine 7 (APC-CY 7) conjugates are supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2- 8 degree C. Do not freeze! ; The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20 degree C. ; The Cyanine 5 (CY 5) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C. ; The Alexa Fluor 488 (AF488) conjugate is supplied as 0.1mg in 0.2mL of PBS/NaN3. Store at 2-8 degree C. ; Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20 degree C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed.

Application Data

(Cell from BALB/c mesenteric lymph nodes were double-stained with Rat Anti-Mouse CD45 (B220) -PE and Rat Anti-mouse CD3 -FITC(clone#C636.29B). Small lymphocytes were then gated and analyzed by flow cytometry.)
