Background: The CD8 antigen is a cell surface glycoprotein found on majority of cytotoxic T lymphocytes, thymocytes and some peripheral blood T cells. It mediates efficient cell-cell interactions within the immune system. CD8 antigen acts as a coreceptor with the T-cell receptor on the T lymphocyte to recognize antigens displayed by an antigen presenting cell in the context of class I MHC molecules. Studies show that different antibodies against human CD8 exert opposite effects on MHC-peptide tetramer binding to the TCR: tetramer staining was enhanced by OKT8 antibody, while it was blocked with SK1 antibody. CD8a is a critical coreceptor of cytotoxic T cells.
